Degradation proceeds along several parallel routes. Deamidation of asparagine and glutamine residues generates charged variants that shift retention time in chromatographic analysis. Oxidation targets methionine and can be accelerated by trace metals or dissolved oxygen. Non-covalent aggregation produces dimers, oligomers, and larger species that are difficult to reverse. Isomerisation at aspartate residues is slower but measurable under thermal stress. The distribution among these pathways depends on pH, buffer composition, ionic strength, and the presence of excipients such as sugars or surfactants.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nm is the standard purity method, reported as area percent. Mass spectrometry, usually with electrospray ionisation, confirms identity and reveals covalent modifications. Size-exclusion chromatography quantifies aggregates and fragments. Peptide mapping after enzymatic digestion localises changes to specific sequence regions. Circular dichroism and infrared spectroscopy report on secondary structure, while light scattering tracks particle formation in liquid formulations. No single technique captures every quality attribute.
Quality control relies on pharmacopoeial monographs where they exist, combined with in-house specifications for identity, purity, water content, and counter-ion composition. Reference standards allow calibration across laboratories, although certified materials for every analogue are not universally obtainable. Batch records, chromatograms, and mass spectra form the documentation trail. Regulatory classification varies by jurisdiction and intended use, and research-grade material differs from pharmaceutical-grade material in testing scope. Analytical uncertainty is often expressed as relative standard deviation across replicate injections.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.

GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.
allele One of multiple alternative versions of an individual gene, each of which is a viable DNA sequence occupying a given position, or locus, on a chromosome. For example, in humans, one allele of the eye-color gene produces blue eyes and another allele of the same gene produces brown eyes.
==== Reaction with lipids ==== Hypochlorous acid reacts with unsaturated bonds in lipids, but not saturated bonds, and the ClO− ion does not participate in this reaction. This reaction occurs by hydrolysis with addition of chlorine to one of the carbons and a hydroxyl to the other. The resulting compound is a chlorohydrin. The polar chlorine disrupts lipid bilayers and could increase permeability. When chlorohydrin formation occurs in lipid bilayers of red blood cells, increased permeability occurs. Disruption could occur if enough chlorohydrin is formed. The addition of preformed chlorohydrin to red blood cells can affect permeability as well. Cholesterol chlorohydrin have also been observed, but do not greatly affect permeability, and it is believed that Cl2 is responsible for this reaction. Hypochlorous acid also reacts with a subclass of glycerophospholipids called plasmalogens, yielding chlorinated fatty aldehydes which are capable of protein modification and may play a role in inflammatory processes such as platelet aggregation and the formation of neutrophil extracellular traps.
Production of the BG Familia mostly ended on 24 May 1994, although the 1.3 L hatchback was kept in production until October 1996 as there was originally no 1.3 L option in the following generation. In Australia and South America, the BG saloon continued to be sold until 1996 as a more affordable alternative to the newer BH model. The Indonesian market 323 was assembled locally starting in 1989 and was available only as a saloon. It was sold in a single trim called Interplay, powered by a carbureted 1.6 L SOHC engine mated to a 5-speed manual or a rare 4-speed automatic transmission. Unique to this market, the 323 came standard with a digital speedometer from the Eunos 100, a feature that was optional in Japan. Around late 1994, a special edition called Interplay Executive was introduced featuring the facelifted bumpers and bigger 14-inch steel wheels with hubcaps, but it also loses the digital speedometer. Also a few months later in this year when the newer BH model was introduced, BG model production was extended until 1997, primarily for the taxi fleet market. This fleet version had its premium features removed and an optional 1.3 L engine. In China, Haima—a joint venture between Hainan Auto Works and Mazda—began manufacturing the BG saloon on the island of Hainan in 1994. The saloon was initially marketed as the HMC6430, powered by a 1.3 L B3 engine. Another model, the GH7160, was released shortly thereafter with a 1.6 L SOHC B6 engine, manufactured at Haima's branch factory in Beihai, Guangxi.
Firearms is a first-person shooter mod for Half-Life which originated from a Quake modification. Initially developed in 1998, Firearms was created as a quasi-realistic team-based FPS. The mod's main feature is the large amount of usable weapons in the game.
Subsequently, some defensins can aggregate to form 'channel-like' pores; others might bind to and cover the microbial membrane in a 'carpet-like' manner. The net outcome is the disruption of membrane integrity and function, which ultimately leads to the lysis of microorganisms. Some defensins are synthesized as propeptides which may be relevant to this process. Alpha defensins of the mouse bowel were historically called cryptdins when first discovered.

As noted above, in radioactive decay the half-life is the length of time after which there is a 50% chance that an atom will have undergone nuclear decay. It varies depending on the atom type and isotope, and is usually determined experimentally. See List of nuclides. The current flowing through an RC circuit or RL circuit decays with a half-life of ln(2)RC or ln(2)L/R, respectively. For this example the term half time tends to be used rather than "half-life", but they mean the same thing. In a chemical reaction, the half-life of a species is the time it takes for the concentration of that substance to fall to half of its initial value. In a first-order reaction the half-life of the reactant is ln(2)/λ, where λ (also denoted as k) is the reaction rate constant.
Once a male is able to retract his foreskin, penile hygiene should become an important feature of his routine body care. Although the American Academy of Pediatrics states there is "little evidence to affirm the association between circumcision status and optimal penile hygiene", various studies suggest that males be educated about the role of hygiene, including retracting the foreskin while urinating and rinsing under it and around the glans at each bathing opportunity. Regular washing under the foreskin was found by Krueger and Osborn (1986) to reduce the risk of numerous penile disorders, however Birley et al. (1993) reports excessive washing with soap should be avoided because it dries the oils out of the tissues and can cause non-specific dermatitis.
=== Separation of powers of the federal and state governments === The federation agreement (Perjanjian Persekutuan) set the powers of the federal and state governments. Financial matters must be handled by the respective states. The Sultan was given full power on religious issues and Malay customs. Foreign policy and defence continued to be administered by the British government. The federation agreement was made the Constitution of the Federation of Malaya and officially declared on 1 February 1948.
=== Abnormal folate metabolism === Several lines of evidence indicate abnormalities of folate metabolism in ASD. These abnormalities can lead to a decrease in 5-methyltetrahydrofolate production, alter the production of folate metabolites and reduce folate transport across the blood-brain barrier and in neurons. The most significant abnormalities of folate metabolism associated with ASD may be autoantibodies to the alpha folate receptor (FRα). These autoantibodies have been associated with cerebral folate deficiency. Autoantibodies can bind to FRα and greatly impair its function. In 2013, one study reported that 60% and 44% of 93 children with ASD were positive for FRα-blocking and binding autoantibodies, respectively. This high rate of anti-FRα autoantibody positivity was confirmed by Ramaekers et al. who compared 75 children with ASD to 30 non-autistic "controls". These controls were children who had a developmental delay, but did not have ASD. FRα-blocking autoantibodies were positive in 47% of children with ASD, but only in 3% of children without ASD. The authors of this study (RE Frye, JM Sequeira, EV Quadros, SJ James and DA Rossignol) have contributed to other such studies on FRα autoantibodies with similar results, however are considered to have a conflict of interest by the publishers. Many children with ASD and cerebral folate deficiency have marked improvements in their clinical status when taking folinic acid.

=== 1963–1998: Bob Rosenberg === In 1963, Rosenberg's son Bob became CEO of the company at age 25, and Dunkin' Donuts opened its hundredth location that year. Dunkin' Donuts was a subsidiary of Universal Food Systems at the time, a conglomerate of 10 small food-service businesses, and Dunkin' Donuts locations varied greatly in their menu options, with some selling full breakfasts and others serving only doughnuts and coffee. In the following years, the other businesses in the Universal Food Systems portfolio were sold or closed, and the company was renamed to Dunkin' Donuts. The menu and shop format was standardized, and various new menu items were introduced. In the early 1980s, counter service and ceramic coffee cups were replaced by self-service counters and paper cups. The chain went public in 1968 and was acquired by Baskin-Robbins owner Allied Lyons in 1990. Dunkin' Donuts expanded in the 1990s by buying out two rival chains: Mister Donut (which was founded by Bill Rosenberg's former partner Harry Winokur) and Dawn Donuts.
The most common dashi stocks for miso soup are made of niboshi (dried baby sardines), kombu (dried kelp), katsuobushi (thin shavings of fermented, dried, and smoked bonito), or hoshi-shiitake (dried shiitake). The kombu can also be used with katsuobushi or hoshi-shiitake. Kelp or shiitake dashi give a vegetarian soup stock. When the soup includes clams such as asari, shijimi, or hamaguri, prepared dashi is often not used. Outside Japan, American- or European-style miso soup is sometimes made by dissolving miso in a Western vegetable stock. It can also be made with chicken broth. The stock might include ingredients such as negi, carrot, potato and daikon radish. In some versions of the dish, chicken stock, Western-style fish stock, and other non-dashi bases can even be used, but there is some debate over whether or not miso soups made using these non-traditional bases count as true miso soup.
The major source of nickel exposure is oral consumption, as nickel is essential to plants. Typical background concentrations of nickel do not exceed 20 ng/m3 in air, 100 mg/kg in soil, 10 mg/kg in vegetation, 10 μg/L in freshwater and 1 μg/L in seawater. Environmental concentrations may be increased by human pollution. For example, nickel-plated faucets may contaminate water and soil; mining and smelting may dump nickel into wastewater; nickel–steel alloy cookware and nickel-pigmented dishes may release nickel into food. Air may be polluted by nickel ore refining and fossil fuel combustion. Humans may absorb nickel directly from tobacco smoke and skin contact with jewelry, shampoos, detergents, and coins. A less common form of chronic exposure is through hemodialysis as traces of nickel ions may be absorbed into the plasma from the chelating action of albumin. The average daily exposure is not a threat to human health. Most nickel absorbed by humans is removed by the kidneys and passed out of the body through urine or is eliminated through the gastrointestinal tract without being absorbed. Nickel is not a cumulative poison, but larger doses or chronic inhalation exposure may be toxic, even carcinogenic, and constitute an occupational hazard. Nickel compounds are classified as human carcinogens based on increased respiratory cancer risks observed in epidemiological studies of sulfidic ore refinery workers. This is supported by the positive results of the NTP bioassays with Ni sub-sulfide and Ni oxide in rats and mice.

Reverse-phase high-performance liquid chromatography with ultraviolet detection is the usual choice, with results reported as area percent. Complementary methods such as size-exclusion chromatography and mass spectrometry are needed because a single separation cannot resolve every impurity class. Purity figures are therefore method dependent and should always be read alongside the technique used.
Oxidation mainly affects methionine residues and is promoted by dissolved oxygen, trace transition metals, and prolonged exposure to light. Buffer choice and the presence of antioxidants in a formulation can alter the rate appreciably. Because the products differ in mass by only a few units, mass spectrometry is often required to detect them.
Published data on long-term ambient stability are limited, so the question remains open rather than settled. Short excursions during transport are common in practice, and many suppliers use insulated packaging with cold packs. Where stability data are absent, cold-chain handling with temperature logging is the safer approach.
It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
