Semaglutide belongs to the glucagon-like peptide-1 receptor agonist class, a group of synthetic peptides that imitate an incretin hormone released by intestinal L cells after food intake. Native GLP-1 circulates for only a few minutes because dipeptidyl peptidase-4 cleaves it rapidly. The hormone acts on pancreatic islets, the gastrointestinal tract, and several brain regions. Because the natural peptide is short-lived, development work concentrated on analogues that keep receptor activity while resisting enzymatic breakdown and renal clearance.
The semaglutide sequence is a 31-residue analogue of human GLP-1, altered at three positions relative to the parent hormone. Aminoisobutyric acid replaces alanine at position 8, arginine replaces lysine at position 34, and a lipophilic diacid is attached to lysine 26 through a short linker. These features are reported consistently in the structural literature. The position 8 substitution blocks recognition by dipeptidyl peptidase-4, while the attached chain drives strong, reversible association with a carrier protein in blood.
Receptor activation raises intracellular cyclic AMP through Gs coupling, which promotes glucose-dependent insulin release and suppresses glucagon secretion when blood glucose is elevated. Effects outside the pancreas include slower gastric emptying and altered appetite signalling in the hypothalamus and hindbrain. The relative contribution of each tissue to overall metabolic outcomes remains an area of active investigation. Central mechanisms in particular are inferred mainly from animal models and indirect human measures rather than direct observation.

Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
Material described as research-grade is not necessarily manufactured to pharmaceutical standards, and purity figures depend on the method used to obtain them. A certificate of analysis states the measured purity, the analytical technique, and the batch identifier, but the underlying data are not always included. Independent testing by a second laboratory is a common way to confirm identity and purity. Uncertainties remain about how storage history affects long-term stability, and about how well results from one laboratory transfer to another. Documentation of handling conditions supports comparison between batches.
Peptides are sensitive to temperature, light, oxygen, and repeated freeze-thaw cycles. Semaglutide in dry form is generally held at refrigerated temperatures, while reconstituted solutions require a defined short-term storage window. Vials should be kept in secondary packaging to limit photodegradation, and exposure to alkaline conditions is avoided because it accelerates chemical degradation. Adsorption to glass and some plastics can reduce the measured concentration of dilute solutions, so low-binding polypropylene containers are preferred for analytical work. Each transfer step introduces a small risk of contamination, and closed handling practices reduce that risk.
A member of the People's Assembly (Arabic: عضو مجلس الشعب) refers to members of the Parliament of Syria. Historically, members of the Syrian parliament have been referred to in Arabic as nā'ib (نائب, "deputy"), plural nuwāb (نواب), a usage rooted in the legislature's earlier incarnation as the Chamber of Deputies (1932–1963). Following the parliament's reconstitution as the People's Assembly under Ba'athist rule in 1971, members came to be more commonly styled "member of the People's Assembly" in official and state-media usage. Nonetheless, "deputy" has persisted as a recognised designation, including in coverage of the current transitional-era Assembly formed after 2024.
Pagoclone is an anxiolytic agent from the cyclopyrrolone family, related to better-known drugs such as the sleeping medication zopiclone. It was synthesized by a French team working for Rhone-Poulenc & Rorer S.A. Pagoclone belongs to the class of nonbenzodiazepines, which have similar effects to the older benzodiazepine group, but with quite different chemical structures. It was never commercialised. It binds with roughly equivalent high affinity (0.7–9.1 nM) to the benzodiazepine binding site of human GABAA receptors containing either an α1, α2, α3 or α5 subunit. It is a partial agonist at α1-, α2- and α5-containing GABAA receptors and a full agonist at receptors containing an α3 subunit. In rats 5′-hydroxypagoclone was identified as a major metabolite. This metabolite has a considerably greater efficacy at the α1 subtype than the parent compound and was shown to have significant anxiolytic-like activity and to produce sedation. In contrast to zopiclone, pagoclone produces anxiolytic effects with little sedative or amnestic actions at low doses (0.3mg to 1.2mg per day). The pharmacologist David Nutt has suggested pagoclone as a possible base from which to make a better social drug, as it produces the positive effects of alcohol, such as relaxation and sociability, but without also causing the negative effects like aggression, amnesia, nausea, loss of coordination and liver damage. Its effect can be quickly reversed by the action of flumazenil, which is already used as an antidote to benzodiazepine overdose.

=== Confirmation === In December 2015, the Joint Working Party of international scientific bodies International Union of Pure and Applied Chemistry (IUPAC) and International Union of Pure and Applied Physics (IUPAP) recognized the element's discovery and assigned the priority of the discovery to the Dubna–Livermore collaboration. This was on account of two 2009 and 2010 confirmations of the properties of the granddaughter of 294Og, 286Fl, at the Lawrence Berkeley National Laboratory, as well as the observation of another consistent decay chain of 294Og by the Dubna group in 2012. The goal of that experiment had been the synthesis of 294Ts via the reaction 249Bk(48Ca,3n), but the short half-life of 249Bk resulted in a significant quantity of the target having decayed to 249Cf, resulting in the synthesis of oganesson instead of tennessine. From 1 October 2015 to 6 April 2016, the Dubna team performed a similar experiment with 48Ca projectiles aimed at a mixed-isotope californium target containing 249Cf, 250Cf, and 251Cf, with the aim of producing the heavier oganesson isotopes 295Og and 296Og. Two beam energies at 252 MeV and 258 MeV were used. Only one atom was seen at the lower beam energy, whose decay chain fitted the previously known one of 294Og (terminating with spontaneous fission of 286Fl), and none were seen at the higher beam energy. The experiment was then halted, as the glue from the sector frames covered the target and blocked evaporation residues from escaping to the detectors.

== Classical era == Because silver was extensively used as a decorative material and an exchange medium, lead deposits came to be worked in Asia Minor from 3000 BC; later, lead deposits were developed in the Aegean and Laurion. These three regions collectively dominated production of mined lead until c. 1200 BC. Beginning c. 2000 BC, the Phoenicians worked deposits in the Iberian peninsula; by 1600 BC, lead mining existed in Cyprus, Greece, and Sardinia. Rome's territorial expansion in Europe and across the Mediterranean, and its development of mining, led to it becoming the greatest producer of lead during the classical era, with an estimated annual output peaking at 80,000 tonnes. Like their predecessors, the Romans obtained lead mostly as a by-product of silver smelting. Lead mining occurred in central Europe, Britain, Balkans, Greece, Anatolia, Hispania, the latter accounting for 40% of world production.
Group 1.4.1 is associated with connective tissue disorders including scleroderma (with scleroderma being the most common disorder with an estimated 8-19% of scleroderma patients having PAH), lupus, mixed connective tissue disease, rheumatoid arthritis, and Sjogren's syndrome. Group 1.4.2 is associated with HIV, group 1.4.3 associated with portal hypertension (commonly due to liver cirrhosis), group 1.4.4 associated with congenital heart disease, and 1.4.5 associated with schistosomiasis. In areas where schistosomiasis is prevalent, PAH due to schistosomiasis is believed to be the most prevalent variant of PAH. It is estimated that 5-8% of people with schistosomiasis with liver fibrosis have PAH. Group 1.5 PAH is the variant of PAH that is responsive to vasodilators (calcium channel blockers). The response to vasodilation by definition includes a decrease in the pulmonary arterial pressure of 10 mmHg or more to less than 40 mmHg without a decline in the cardiac output upon infusion of nitrous oxide(vasodilator) during the right heart catheterization. Vasodilator responsive PAH is estimated to be 6.8% of PAH patients in one study, and it is associated with a more favorable prognosis. Group 1.6 PAH includes PAH with features of venous or capillary involvement and is thought to comprise 5-10% of PAH cases. Two conditions, pulmonary veno-occlusive disease and pulmonary capillary hemangiomatosis comprise this category. The subcategory is characterized by severe hypoxemia, capillary congestion and prominent post-capillary venule thickening.
Inadequate nutritional intake: Alcoholics tend to take in less than the recommended amount of thiamine. Decreased uptake of thiamine from the GI tract: Active transport of thiamine into enterocytes is disturbed during acute alcohol exposure. Liver thiamine stores are reduced due to hepatic steatosis or fibrosis. Impaired thiamine utilization: Magnesium, which is required for the binding of thiamine to thiamine-using enzymes within the cell, is also deficient due to chronic alcohol consumption. The inefficient use of any thiamine that does reach the cells will further exacerbate the thiamine deficiency. Ethanol per se inhibits thiamine transport in the gastrointestinal system and blocks phosphorylation of thiamine to its cofactor form (ThDP). Following improved nutrition and the removal of alcohol consumption, some impairments linked with thiamine deficiency are reversed, in particular poor brain functionality, although in more severe cases, Wernicke–Korsakoff syndrome leaves permanent damage. From a clinical perspective, two significant school of thought exists for psychiatric conditions associated with cannabis (or cannabinoids) use: transient, non-persistent psychotic reactions, and longer-lasting, persistent disorders that resemble schizophrenia. The former is formally known as acute cannabis-associated psychotic symptoms (CAPS) or cannabis-induced psychotic disorder (CIPD). At an epidemiological level, a dose–response relationship exists between cannabis use and increased risk of psychosis and earlier onset of psychosis. Although the epidemiological association is robust, evidence to prove a causal relationship is lacking. Cannabis may also increase the risk of depression, but insufficient research has been performed to draw a conclusion. A 2014 meta-analysis of longitudinal studies found that cannabis use was associated with a modestly increased risk of developing depressive disorders, particularly among heavy users, although the causal relationship remains uncertain and further research is needed. Cannabis use is associated with increased risk of anxiety disorders, although causality has not been established. A 2025 systematic review and meta-analysis involving more than half a million participants aged 15–30 reported that cannabis use was associated with higher odds of depression (51% higher), anxiety (58%), suicidal ideation (65%), and suicide attempt (87%). A review in 2019 found that research was insufficient to determine the safety and efficacy of using cannabis to treat schizophrenia, psychosis, or other mental disorders.
=== Unhealthy weight loss === Some unhealthy weight loss illnesses can include cancer, diseases such as liver and kidney disease, diabetes, hyperthyroidism, and eating disorders such as anorexia and bulimia. It is important to know the difference between healthy weight loss and unhealthy weight loss.
The ATP synthase of mitochondria and chloroplasts is an anabolic enzyme that harnesses the energy of a transmembrane proton gradient as an energy source for adding an inorganic phosphate group to a molecule of adenosine diphosphate (ADP) to form a molecule of adenosine triphosphate (ATP). This enzyme works when a proton moves down the concentration gradient, giving the enzyme a spinning motion. This unique spinning motion bonds ADP and P together to create ATP. ATP synthase can also function in reverse, that is, use energy released by ATP hydrolysis to pump protons against their electrochemical gradient.

== Epidemiology == Globally, the prevalence and incidence of pulmonary fibrosis has been studied in the United States, Norway, Czech Republic, Greece, United Kingdom, Finland, and Turkey, with only two studies in Japan and Taiwan. But most of these studies were of people already diagnosed with pulmonary fibrosis, which lowers the diagnosis sensitivity, so that the prevalence and incidence has ranged from 0.7 per 100,000 in Taiwan to 63.0 per 100,000 in the U.S., and the published incidence has ranged from 0.6 per 100,000 person years to 17.4 per 100,000 person years. The mean age of all pulmonary fibrosis patients is between 65 and 70 years, making age a criterion of its own. Aging respiratory systems are much more vulnerable to fibrosis and stem cell depletion.

It is a synthetic peptide of 31 amino acids, built to resemble the natural incretin hormone GLP-1. Because of its size and composition it is handled analytically like other therapeutic peptides, using chromatographic and mass spectrometric methods rather than the techniques typical of small organic drugs.
