Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.

== Prevention of nerve injuries == Methods to help prevent nerve injuries include injection pressure monitoring. The presence of a high opening injection pressure (> 20 PSI) is a sensitive sign of intrafascicular/intraneural needle tip placement. Extrafascicular needle tip placement is associated with low pressures (< 20 PSI). Also, high pressure injection was associated with neurologic deficits and severe axonal damage after the block. Other methods of preventing nerve injury include electrical nerve stimulation and ultrasonography. Electrical stimulation with a motor response at < 0.2 mA only can occur with an intraneural/intrafasciular needle tip location.

The units use strontium-90, and a larger number of such units have been deployed both on the ground and on the ocean floor than have been used on spacecraft, with public regulatory documents suggesting that the US had deployed at least 100–150 during the 1970s and 1980s. In the past, small "plutonium cells" (very small 238Pu-powered RTGs) were used in implanted heart pacemakers to ensure a very long "battery life". As of 2004, about ninety were still in use. By the end of 2007, the number was reported to be down to just nine. The Mound Laboratory Cardiac Pacemaker program began on 1 June 1966, in conjunction with NUMEC. The program was cancelled in 1972, when it was recognized that the heat source would not remain intact during cremation and there was no way to completely ensure that the units would not be cremated with their users' bodies.
== History == Cyclic guanosine monophosphate (cGMP) research began after cGMP and cyclic adenosine monophosphate (cAMP) were identified as cellular components and potentially involved with cellular regulation. Upon the synthesis of cGMP in 1960, progress rapidly spread in the understanding of regulation and effects of cGMP. Earl W. Sutherland received the 1971 Nobel Prize in Medicine for his work with cAMP and secondary messengers. This award sparked extensive research into cAMP, while cGMP received less attention, with its biological functions largely unknown until the 1980s. During this period, two pivotal discoveries highlighted cGMP's role in cellular signaling: atrial natriuretic peptide (ANP) was found to stimulate cGMP synthesis through the particulate guanylyl cyclase (pGC) receptor, and nitric oxide (NO), identified as the endothelium-derived relaxing factor, was shown to activate soluble guanylyl cyclase (sGC), producing cGMP to mediate vasodilation in smooth muscle cells. Further components involved with the cGMP were also identified such as cGMP-hydrolyzing phosphodiesterases (PDEs) and cGMP-binding proteins. The awarding of the 1998 Nobel Prize to Robert F. Furchgott, Louis J. Ignarro, and Ferid Murad for their discoveries in the NO-cGMP pathway renewed interest in cGMP research with the 1st International Conference on cGMP being held in 2003.

Adrenomedullin (ADM) is a multifunctional peptide hormone that plays an important role in the homeostasis of the cardiovascular system and in inflammatory response. It acts as a potent vasodilator, regulating vascular tone and blood pressure through both endothelium-dependent and independent mechanisms. ADM exerts protective effects on the cardiovascular system by inhibiting apoptosis in endothelial cells, reducing oxidative stress, and regulating vascular smooth muscle cell proliferation. In the heart, it increases cardiac output and augments myocardial contractility. Beyond its cardiovascular functions, ADM demonstrates significant anti-inflammatory properties, modulating cytokine production and secretion in macrophages. It also contributes to the maintenance of vascular integrity, potentially reducing vascular permeability during inflammatory conditions. In addition, ADM has been implicated in angiogenesis, protection of organs, and tissue repair. Because of its wide-ranging effects, it has potential therapeutic applications in a variety of diseases, including inflammatory bowel disease, sepsis, and cardiovascular disorders.
An – actinide Some actinides can exist in several oxide forms such as An2O3, AnO2, An2O5 and AnO3. For all actinides, oxides AnO3 are amphoteric and An2O3, AnO2 and An2O5 are basic, they easily react with water, forming bases:
On 27 February at 3:38 p.m. EST (11:08 p.m. IRST), Trump, traveling on Air Force One to Texas, authorized Operation Epic Fury. US missiles, drones, and Israeli fighter jets began striking Iran the next day, around 9:45 am. IRST (1:15 am. EST). The strikes took place during negotiations over Iran's nuclear program, and coincided with the holy month of Ramadan. The operation was codenamed Operation Roaring Lion by Israel. The Israeli Air Force (IAF) said it struck 500 military targets in Iran in the largest combat sortie in its history. Iranian naval vessels were also targeted. Israel said it used over 1,200 bombs in 24 hours. US strikes were carried out by planes based around the Middle East and from aircraft carriers.

Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
The fatty diacid chain associates strongly with serum albumin, which keeps the peptide in circulation and shields it from rapid renal clearance. Bound peptide is released gradually, producing a sustained receptor signal. This mechanism also reduces the peak-to-trough variation seen with shorter-acting analogues.
The balance between peripheral receptor activation and signalling in the central nervous system is not fully resolved. The extent to which slowed gastric emptying accounts for reduced energy intake, compared with direct effects on appetite circuits, is debated. Long-term effects on lean mass and on tissues outside the gut and brain are still under study.
冻融过程中冰晶形成与局部浓度升高会促使肽链发生界面吸附和聚集。聚集不仅降低有效含量,还会改变可见异物与不溶性微粒的计数结果。将溶液分装为单次使用的小体积等份可减少循环次数。
