In trying to find a way to defeat GLaDOS, he finds that Chell, one of the human subjects kept in cryogenic storage within Aperture, has a high level of tenacity, and arranges for the events of Portal to occur by moving her to the top of GLaDOS' testing list. GLaDOS remains driven to test human subjects despite the lack of humans.
=== Inhalable insulin === Inhalable insulin, also known as technosphere insulin, is a powdered form of insulin, delivered with an inhaler into the lungs where it is absorbed. In general, inhaled insulins have been more rapidly absorbed than subcutaneous injected insulin, with faster peak concentration in serum and more rapid metabolism. Exubera, developed by Inhale Therapeutics (later named Nektar Therapeutics), became the first inhaled insulin product to be marketed, in 2006 by Pfizer, but poor sales led Pfizer to withdraw it in 2007. Afrezza, a monomeric inhaled, ultra rapid-acting insulin developed by Mannkind, was approved by the FDA in 2014 and is the only inhaled insulin commercialized as of 2025.
=== Regulation === In the United States, the Food and Drug Administration (FDA) regulates nicotine as a tobacco product under the 2009 Family Smoking Prevention and Tobacco Control Act. The FDA is tasked with reviewing tobacco products such as e-cigarettes and determining which can be authorized for sale. In March 2022, the US Congress passed a law (the Consolidated Appropriations Act, 2022) that expanded FDA's tobacco regulatory authority to include tobacco products containing nicotine from any source, thereby including products made with synthetic nicotine. On January 17, 2025, the FDA proposed a strongly endorsed rule to reduce nicotine in cigarettes and certain combusted tobacco products to minimally or non-addictive levels, capping nicotine yield at 0.7 mg per gram of tobacco, approximately a 95% reduction from current commercial levels. In the European Union, the Tobacco Products Directive (2014/40/EU) regulates the manufacture, presentation, and sale of tobacco and related products. In the United Kingdom, the Tobacco and Related Products Regulations 2016 implemented the European directive 2014/40/EU, amended by Tobacco Products and Nicotine Inhaling Products (Amendment etc.) (EU Exit) Regulations 2019 and the Tobacco Products and Nicotine Inhaling Products (Amendment) (EU Exit) Regulations 2020. == Medical use == According to phase III randomized trials, a cangrelor–clopidogrel combination is safe and has been found to be more effective than standard clopidogrel treatment at reducing ischemic events in the heart, without increasing major bleeding in the treatment of stenotic coronary arteries. The advantages of this drug combination are most prominent in patients with myocardial infarction. Available antiplatelet drugs have delayed onset and offset of action. Since cangrelor's effects are immediate and quickly reversed, it is a more desirable drug for elective treatment of stenotic coronary arteries, high risk acute coronary syndromes treated with immediate coronary stenting, and for bridging those surgery patients who require P2Y12 inhibition. Evidence regarding cangrelor therapy is limited by the lack studies assessing cangrelor administration in conjunction with either prasugrel or ticagrelor. Cangrelor been approved for adults undergoing percutaneous coronary intervention (PCI).

In October, Clarke arrived at Wepener at the head of a force of 1,000 cavalry, 600 infantry, five artillery pieces, and 40 wagons. Clarke was aiming to relieve Mafeteng, whose garrison was forced to exchange messages written in Greek, since some Basuto chiefs spoke both English and French. Clarke's advance was slowed by deep mud, and the Basuto cavalry regularly harried the column with rifle fire before withdrawing. On 19 October, the Cape army reached Qalabane, an isolated kop halfway between Wepener and Mafeteng. Lerotholi had positioned 3,000 of his warriors behind a ridge that overlooked a nearby road. The advanced guard of the Cape Mounted Yeomanry came under rifle fire from the kop. Clarke ordered the artillery to fire upon the kop and dispatched 200 men from the 1st Cape Mounted Yeomanry to flank the kop from the left. CMY commander Captain Dalgety ordered his soldiers to dismount and assume an open order formation. Chief Seiso's led a charge of 300 axe-wielding Basuto cavalrymen on Dalgety's unit before the latter was able to reach the crest. The 2nd CMY reinforced Dalgety soon afterwards and captured a nearby village. The Cape army lost 32 killed and seven injured, while the Basuto lost 40 killed. The yeomanry was almost defenseless in hand-to-hand combat, as it was not yet issued bayonets or swords.

Bromantane, sold under the brand name Ladasten, is an atypical central nervous system (CNS) stimulant and anxiolytic drug of the adamantane family that is related to amantadine and memantine. Medically, it is approved in Russia for the treatment of neurasthenia. Although the effects of bromantane have been determined to be dependent on the dopaminergic and possibly serotonergic neurotransmitter systems, its exact mechanism of action is unknown, and is distinct in its properties relative to typical stimulants such as amphetamine. It is performed via timing a jump the exact moment you land on the ground, doing so make's the engine preserve your current velocity. This exploit and also be paired with something called Air-Strafing to gain speed while the mid-air.
=== Indole butyrate disk === An indole butyrate disc is used to differentiate between Neisseria gonorrhoeae (negative result) and Moraxella catarrhalis (positive result). This test involves a butyrate disk, which when smeared with a culture, will change color for a positive result after 5 minutes of incubation. A blue color is the result of a positive test.
Around 99.8% of genes that undergo mutations are deemed silent because the nucleotide change does not change the amino acid being translated. Although silent mutations are not supposed to have an effect on the phenotypic outcome, some mutations prove otherwise like the Multi-Drug Resistance Gene 1. MDR1 codes for the P-glycoprotein which helps get rid of drugs in the body. It is located in the intestines, liver, pancreas, and brain. MDR 1 is located in the same places that CYP3A4 is located in, which is an enzyme that helps get rid of toxins or drugs from the liver and intestines. Silent mutations like MDR 1 do express a change in phenotypic response. A study done on mice showed when they did not have enough of the MDR 1 gene, their body did not recognize the ivermectin or cyclosporine drug, leading to the creation of toxins in their bodies. MDR1 has over fifty single nucleotide polymorphisms (SNP's) which are changes in the nucleotide base sequence. In MDR1 the gene exon 26 which represents 3535C can mutate to 3535T which then changes the transfer RNA into one that is not often as seen, leading to changes in the outcome during translation. This is an example of how some silent mutations are not always silent. The multi-drug resistance genes at Exon 26 C3435T, exon 21 G2677T/A, and exon 12 C1236T have been studied to have SNP's that occur at the same time, therefore making the phenotypic "function "change. This suggests a haplotype dependency between exon 26 and other exon that have polymorphisms.
