Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
Storage conditions for semaglutide depend heavily on the presentation. Lyophilized research powder is generally kept at two to eight degrees Celsius in a sealed container, protected from light and moisture. Manufacturer labeling for finished injectable products specifies refrigeration before first use, with defined in-use periods at room temperature afterward. The oral tablet form is stored at controlled room temperature and is more tolerant of short excursions. Temperature excursions should be documented rather than inferred.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
Identity and purity are usually assessed by reverse-phase high-performance liquid chromatography coupled to mass spectrometry. Retention time and observed mass are compared against a reference standard run under identical conditions. Impurity profiles reveal deamidation products, oxidized methionine variants, and truncated fragments that arise during synthesis or storage. Peptide mapping through enzymatic digestion confirms the primary sequence, while amino acid analysis offers an independent check on overall composition.

GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
== History == EAS was founded in Pacific Grove, Monterey County, California in 1992 by biochemist Anthony Almada, and businessman Ed Byrd. In 2012, Byrd founded EAB Lab. After discovering the possible benefits for athletes of creatine monohydrate supplementation, in December 1992 they acquired a kilogram of creatine and began trials. In 1993, they released the first commercially available creatine supplement intended specifically for strength and muscle enhancement under the EAS brand name Phosphagain. The product caught the eye of Bill Phillips, publisher of Muscle Media 2000 (usually referred to as "MM2K"), and he began to write about its benefits in the Natural Supplement Review and his subscriber-based newsletters. In 1994 he purchased EAS from Almada and Byrd, and was 100% owner of EAS until the sale to North Castle Partners. Phillips' subscriber base was the platform in which to tout his new acquisition. He promoted the company's products through heavy editorial-style advertisements in MM2K. Led by products like Myoplex Protein Supplement, Phosphagen and HMB, this eventually made EAS significant in the nutritional supplement industry. In 1999, North Castle Partners purchased EAS for $160 million. Phillips retained about a third of the company, which he sold in 2004. In 2005, EAS was purchased by Abbott Laboratories for approximately $320 million in a cash-for-stock transaction and existed as a solely-owned division of Abbott. The refreshed product line focused on sports bars, drink mixes, and similar products.
The antibody, together with the captured target peptide, is then separated from the complex sample digest, after which the highly purified peptide is eluted from the antibody and delivered to a mass spectrometer for measurement. The capture step has been implemented using antibodies bound to magnetic beads as well as antibodies immobilized on flow-through columns. Addition of this specific capture step provides two primary advantages in comparison with a conventional workflow analyzing an unfractionated sample digest: sensitivity and throughput. The antibody can be used to capture the target peptide (and SIS) from a much larger mass of sample than could be analyzed directly by MS, thus allowing lower concentrations to be measured. In practice, assay sensitivity can be improved by 1,000-10,000-fold by this approach. By removing the unbound (non-target) peptides present in the sample digest, the sample presented to the mass spectrometer is drastically simplified, thus reducing the need for peptide separation by liquid chromatography prior to MS analysis. In some cases liquid chromatography has been eliminated entirely, resulting in MS cycle times of 7-20 sec rather than 5–40 minutes required in typical unfractionated digest protocols involving extensive chromatographic separation. By virtue of the extreme specificity of mass spectrometric detection, SISCAPA assays can be combined into multiplex panels without cross-assay interference. Panels combining 22, 50, and 150 assays into a single operation have been demonstrated.
The brisket muscles are sometimes separated for retail cutting: the lean "first cut" or "flat cut" is the deep pectoral, while the fattier "second cut", "point", "fat end", or "triangular cut" is the superficial pectoral. For food service use, they are IMPS 120A and 120B, respectively.
A myocardial infarction is usually clinically classified as an ST-elevation MI (STEMI) or a non-ST elevation MI (NSTEMI). These are based on ST elevation, a portion of a heartbeat graphically recorded on an ECG. STEMIs make up about 25–40% of myocardial infarctions. A more explicit classification system, based on international consensus in 2026, also exists. This classifies myocardial infarctions into three types:
The authors hypothesized that the higher levels of CD8+ T cell infiltration was due to anti-CTLA-4 inhibited the conversion of CD4 T cells to T regulator cells and further reduced T regulatory suppression with anti-PD-1. This combination promoted a more robust inflammatory response to the tumor that reduced the size of the cancer. Most recently, the FDA has approved a combination therapy with both anti-CTLA4 (ipilimumab) and anti-PD1 (nivolumab) in October 2015. The molecular factors and receptors necessary making a tumor receptive to anti-PD1 treatment remains unknown. PD-L1 expression on the surface on cancer cells plays a significant role. PD-L1 positive tumors were twice as likely to respond to combination treatment. However patients with PD-L1 negative tumors also have limited response to anti-PD1, demonstrating that PD-L1 expression is not an absolute determinant of the effectiveness of therapy. Higher mutational burden in the tumor is correlated with a greater effect of the anti-PD-1 treatment. In clinical trials, patients who benefited from anti-PD1 treatment had cancers, such as melanoma, bladder cancer, and gastric cancer, that had a median higher average number of mutations than the patients who did not respond to the therapy. However, the correlation between higher tumor burden and the clinical effectiveness of PD-1 immune blockade is still uncertain. The 2018 Nobel Prize for Medicine was awarded to James P Allison and Tasuku Honjo "for their discovery of cancer therapy by inhibition of negative immune regulation".

== Procedure Steps == For anorectal fistulae, the surgeon begins by identifying the internal and external opening of the fistula. The external opening is usually on the patient's skin and can be identified in clinic. The internal opening is within the anus, and can be found while the surgeon examines the anorectal columns while the patient is under anesthesia. A methylene blue dye or a peroxide solution may be used to aid with this process. Once the openings of the tract are found, a thin metal probe is strung through. Using scissors or electrocautery, the surgeon then cores out the tunneling tract. At this point, the surgeon inspects the anal sphincters and closes any defects that were made during the procedure. The wound can then be left open to heal or closed by approximating the tissue back to its anatomic position with sutures.
Therapeutic monoclonal antibodies (mAbs) are used for several diseases, including cancer and Rheumatoid arthritis. Consequently, the high immunogenicity limited efficacy and was associated with severe infusion reactions. Although the exact mechanism is unclear, it is suspected that the mAbs are inducing infusion reactions by eliciting antibody antigen interactions, such as increased formation of immunoglobulin E (IgE) antibodies, which may bind onto mast cells and subsequent degranulation, causing allergy-like symptoms as well as the release of additional cytokines. Several innovations in genetic engineering has resulted in the decrease in immunogenicity, (also known as deimmunization), of mAbs. Genetic engineering has led to the generation of humanized and chimeric antibodies, by exchanging the murine constant and complementary regions of the immunoglobulin chains with the human counterparts. Although this has reduced the sometimes extreme immunogenicity associated with murine mAbs, the anticipation that all fully human mAbs would have not possess unwanted immunogenic properties remains unfulfilled.
=== Middle Ages === After the fall of the Western Roman Empire, the town, along with the rest of the Italian peninsula, was conquered by the Heruli and the Ostrogoths, recaptured by the Romans, but then conquered again by the Lombards whose territory then fell into the hands of the Franks under Charlemagne (773). The Contea di Torino (countship) was founded in the 940s and was held by the Arduinic dynasty until 1050. After the marriage of Adelaide of Susa with Humbert Biancamano's son Otto, the family of the Counts of Savoy gained control. While the title of count was held by the Bishop as count of Turin (1092–1130 and 1136–1191) it was ruled as a prince-bishopric by the Bishops. In 1230–1235, it was a lordship under the Marquess of Montferrat, styled Lord of Turin. At the end of the 13th century, when it was annexed to the Duchy of Savoy, the city already had 20,000 inhabitants. Many of the gardens and palaces were built in the 15th century when the city was redesigned. The University of Turin was also founded during this period.
=== 14 December === Three explosions were heard in the centre of Kyiv; President Zelenskyy claimed that Ukrainian air defence forces shot down 13 Shahed-136 drones. Reports emerged that U.S. officials were finalising and preparing to announce a plan to provide Ukraine with the sophisticated Patriot air defence system, agreeing to an urgent request from Ukrainian leaders amid increasing Russian missile attacks against Ukraine's infrastructure. The Biden administration was reluctant to deploy the system for months, as a Patriot battery complex would need at least 90 trained troops to operate and maintain it, along with concerns that it would provoke Russia to escalate. The Ukrainian Parliament's Commissioner for Human Rights Dmytro Lubinets claimed that a children's torture chamber had been uncovered in Kherson. Andrii Yermak, Chief of Staff of the Office of President Zelenskyy, stated that they released around 64 military personnel and a US citizen during a prisoner swap deal with Russia.

== Structure and role == The pancreas serves multiple roles within mammalian organisms. It plays a role in the digestive system and the endocrine system making it an organ and a very important gland. PP cells tend to located in the pancreatic islets, and are one of the rarer pancreatic cell types. Some small clusters may also be found surrounded by exocrine pancreas. They tend to be located in the duodenal part of the pancreas more medially. Given their location in pancreatic islets along with their low population, they are able to respond efficiently to changes in blood sugar, nerve impulses, and chemical signaling through gap junctions or cellular signaling. PP cells show the highest concentration at the head of the pancreas. PP cells play an important role when it comes to the regulation of plasma glucose as they help to synthesize and release pancreatic polypeptide (PP). As PP cells secrete more pancreatic polypeptide, it has been shown to have a negative effect on insulin secretion giving it anorexic effects on intestinal functionality. These cells have been shown to inhibit functions in the gut and decrease intestinal motor activity as well. In other words, as the secretion of pancreatic polypeptide increases, the secretion of insulin, a peptide hormone, starts to decrease. Pancreatic polypeptide also works to regulate glycogen levels and the amount of secreted substances from gastrointestinal cells and tissue. Within PP cells and its nucleus is the cellular DNA that allows for different proteins to be made that go on to play a role in cellular functions.

== Career == Crampton began making music under the name E+E ("And & And" in Spanish) in the early 2000s. E+E consisted of several performers and contributing writers, editors, and DJ mixes made with a keyboard, acapellas, and a sampler. In 2015, they ceased using the E+E alias and released their first studio album, American Drift, under the name Elysia Crampton. The album took three years to make and was made as a way to describe their unique experience of finding a home in Virginia in the aforementioned years. The album was released on August 7, 2015, and was met with critical success. The music review website Pitchfork gave the album an 8.1 out of 10 and said, in praise:
Paracetamol poisoning, also known as acetaminophen poisoning, is caused by excessive use of the medication paracetamol (acetaminophen). Most people have few or non-specific symptoms in the first 24 hours following overdose. These symptoms include feeling tired, abdominal pain, or nausea. This is typically followed by absence of symptoms for a couple of days, after which yellowish skin, blood clotting problems, and confusion occurs as a result of liver failure. Additional complications may include kidney failure, pancreatitis, low blood sugar, and lactic acidosis. If death does not occur, people tend to recover fully over a couple of weeks. Without treatment, death from toxicity occurs 4 to 18 days later. Paracetamol poisoning can occur accidentally or as a suicide attempt. Risk factors for toxicity include alcoholism, malnutrition, and the taking of certain other hepatotoxic medications. Liver damage results not from paracetamol itself, but from one of its metabolites, N-acetyl-p-benzoquinone imine (NAPQI). NAPQI decreases the liver's glutathione and directly damages cells in the liver. Diagnosis is based on the blood level of paracetamol at specific times after the medication was taken. These values are often plotted on the Rumack-Matthew nomogram to determine level of concern. Treatment may include activated charcoal if the person seeks medical help soon after the overdose. Attempting to force the person to vomit is not recommended. If there is a potential for toxicity, the antidote acetylcysteine is recommended. The medication is generally given for at least 24 hours.

Sealed, protected from light, and refrigerated at two to eight degrees Celsius for most research material. Desiccated storage limits moisture uptake. Allow the vial to reach room temperature before opening to prevent condensation.
Each cycle exposes the peptide to ice-liquid interfaces where unfolding and aggregation can occur. Aggregates may not redissolve and can alter measured activity. Aliquoting before the first freeze reduces the number of cycles any single portion experiences.
It usually reports purity by HPLC, identity by mass spectrometry, peptide content, appearance, and residual moisture or counterion content. The analytical method behind each value should be stated. Acceptance criteria are set by the supplier or the buyer's specification.
Manufacturers commonly state multi-year stability when the powder is kept dry and frozen. Actual shelf life depends on residual moisture, vial sealing, and storage temperature. A re-test by chromatography is the only way to confirm remaining purity.
