N-Acetylcysteine ethyl ester (NACET, NAC-Et) is an antioxidant which acts as a lipophilic prodrug for N-acetylcysteine and subsequently glutathione, with improved bioavailability over N-acetylcysteine itself, and has been researched in animal studies as a potential treatment for diabetic retinopathy. It has also been sold as a "dietary supplement" in combination with glycine under the name GlyNAC-Et as a supposedly improved version of the older glycine-NAC combination GlyNAC which has been claimed to show efficacy in counteracting symptoms associated with aging as well as HIV infection, diabetes and nerve damage. == Amino-acid preferences == One might expect that larger side chains would result in more restrictions and consequently a smaller allowable region in the Ramachandran plot, but the effect of side chains is small. In practice, the major effect seen is that of the presence or absence of the methylene group at Cβ. Glycine has only a hydrogen atom for its side chain, with a much smaller van der Waals radius than the CH3, CH2, or CH group that starts the side chain of all other amino acids. Hence it is least restricted, and this is apparent in the Ramachandran plot for glycine (see Gly plot in gallery) for which the allowable area is considerably larger. In contrast, the Ramachandran plot for proline, with its 5-membered-ring side chain connecting Cα to backbone N, shows a limited number of possible combinations of ψ and φ (see Pro plot in gallery). == Other epitope-tag combinations == The principle of tandem-affinity purification of multiprotein complexes is not limited to the combination of CBP and Protein A tags used in the original work by Rigaut et al. (1999). For example, the combination of FLAG- and HA-tags has been used since 2000 by the group of Nakatani to purify numerous protein complexes from mammalian cells. Many other tag combinations have been proposed since the TAP principle was published.
